In this webinar, we will discuss the presentation, radiological appearance, histomolecular pathology, and management of PLGG from rare subtypes and harboring rare molecular alterations.

Statement of Need/Program Overview

The treatment paradigm for IDH-mutant glioma is evolving with the recent FDA approval of vorasidenib for the treatment of IDH-mutant glioma. While the inclusion criteria for the landmark INDIGO trial was limited to low grade IDH-mutant gliomas (WHO grade 2 only), tumor grading based on histological criteria in adult-type diffuse glioma remains controversial in the current molecular era. This virtual tumor board aims to provide a nuanced discussion of the management of IDH-mutant glioma to illustrate the spectrum of considerations that can arise in clinical practice. This session will review the role of pathological grading and prognostication using molecular markers in IDH-mutant glioma. The current standard of care for IDH-mutant glioma will be discussed, eliciting debate on future directions in research and clinical trials to answer outstanding questions stemming from the INDIGO study in the post-vorasidenib FDA approval era.

Medulloblastoma arising in the AYA population harbor unique molecular characteristic and present a therapeutic challenge to oncologists caring for these patients. The majority of treatments are based on pediatric trials, and there is a lack of a universally accepted standard-of-care treatment approach in the AYA population. Additionally, this population faces distinct challenges in terms of acute and long-term toxicities as well as specific psychosocial needs. A session focused on AYAs with medulloblastoma can deepen our understanding, enhance patient care, and underscore the necessity for future optimized treatment strategies.

Statement of Need/Program Overview

The increasing refinement of CNS tumor classification based on molecular alterations, as well as the emergence of novel therapies targeting these alterations, is heralding the era of precision medicine in neuro-oncology. This SNO Adult Molecular Tumor Board will review cases highlighting the importance of molecular testing in glioma and discuss the use of targeted therapies to effectively treat molecularly-defined tumor types. Real world clinical cases of patients with MAPK pathway and H3 K27 altered gliomas will be presented and discussed by national and international neuro-oncology experts.

Infant-type hemispheric glioma is a new category of pediatric high-grade glioma (HGG) as of the 2021 update to the WHO classification of CNS tumors. These tumors are biologically distinct from HGG in older children in terms of molecular drivers, and they also have more favorable outcomes in many cases. However, management of these tumors in infants presents unique challenges as well. In this webinar, we intend to review and discuss current understanding of the molecular characteristics, subgrouping, treatment challenges, and therapeutic approaches to infant HGG.

Nomenclature related to neurofibromatosis syndromes has recently changed from type 1 and type 2 to neurofibromatosis type 1 and NF2-related schwannomatosis. Along with these changing terms, management of NF-related inherited syndromes and their associated tumors has advanced to include targeted treatment and new strategies. In this webinar, we will discuss the major tumor types associated with both conditions and how management of tumors and these syndromes as a whole has advanced, through a variety of case presentations and an expert panel discussion.
